Abstract

Abstract
Background:
Chemotherapy is regarded as one of the main causes of male infertility, with cyclophosphamide (CP) being a chemotherapy with significant extended drawbacks on male fertility. As such, it has been essential to introduce an effective therapeutic strategy to preserve male fertility. Bone marrow derived mesenchymal stem cells (BM-MSCs) represent a promising therapeutic regimen in a variety of health conditions. In this context, the present study aimed to assess the therapeutic potential of locally injected BM-MSCs on CP induced changes on seminiferous tubules of adult rats.
Materials and methods:
The study design included 38 adult rats, as follows; control group (subgroup IA, received single intraperitoneal (IP) injection of 0.5 ml of sterile saline, IB rats were operated upon and testes were injected with 1 ml of complete culture media and IC represented negative control subgroup). Group II; received single dose of CP (20 mg/kg body weight) in saline and sacrificed after 4 weeks. After four weeks from CP administration, group III rats received locally injected BM-MSCs, while group IV rats represented spontaneous recovery. Groups III and IV were sacrificed after another four weeks. Assessment was performed histologically by light microscopic examination of hematoxylin and eosin (H&E) stained sections and by electron microscopic examination. Immunohistochemical assessment of Bcl-2 was also performed. Morphometric analysis was done for assessment of height of seminiferous epithelium and percentage area of positive reaction to anti- Bcl-2.
Results:
CP induced degenerative changes in spermatogenic cells and reduced height of the seminiferous epithelium. Marked amelioration was observed following BM-MSCs administration with restoration of normal arrangement of the germinal epithelium. Morphometric results revealed significantly increased height of seminiferous epithelium and Bcl-2 expression in BM-MSCs group.
Conclusion:
Local injection of BM-MSCs induced a potent therapeutic effect in restoration of histological architecture of the seminiferous tubules and consequently spermatogenesis.
